As a native Spanish speaker I can tell you with certainty that "pene" is a masculine word, because it ends with "e"

As a native Spanish speaker you should know the gender of nouns that end in "e" can go both ways.

If you want a femenine word for the penis use "la polla". It's more akin to dick, but at least is femenine
